Tissue tolerance and systemic toxicity of 5-fluorouracil, external radiation, and drug-radiation combination in rabbits

      Abstract

      The controversy concerning the true nature of atypical cells circulating in the bloodstream in patients undergoing operations for malignancy and the current status of Thio-TEPA as an adjuvant to radical mastectomy in the cure of breast carcinoma are discussed in the introduction.
      The hypothesis that 5-fluorouracil sensitizes the skin in the radical mastectomy site to subsequent external radiation has been tested in the rabbit and subsequently observed in 17 patients with carcinoma of the breast.
      Both the laboratory and clinical material suggest that the antitumor agent has no effect on the skin when administered prior to external radiation and that there is no apparent contraindication to the use of a combined cancer chemotherapy-radiation program in the treatment of breast carcinoma.
      The clinical results of this combined program are being studied on a long-term basis.
